Амброзиус Бенсон родился на севере Италии в Ломбардии. В 1518 Бенсон приехал в Брюгге, где скоро зарекомендовал себя отличным портретистом и мастером алтарной живописи и был принят в городскую Гильдию художников.
Одно время Бенсон работал с известным мастером Герардом Давидом, оказавшим на его манеру значительное влияние. Однако после нескольких месяцев совместной работы художники расстались, не найдя взаимопонимания.

Бенсон был благополучным и удачливым человеком. Художник не раз исполнял заказы городских властей: по заказу Магистратуры им было расписано помещение счётной палаты. Трижды он избирался в Городской совет, а также занимал важные посты в Гильдии художников: исполнял обязанности декана (1537-1538 и 1543-1544) и управляющего (1540-1541). До самой своей кончины в 1550 году он не покидал Брюгге.
